24iProof polymerase demonstrates unrivaled speed, leading to dramatically shorter overall reaction times. The reactionprotocol for iProof polymerase was compared to the recommended protocols for two competing polymerases. Each protocol wasdesigned to amplify 1, 8, or 15 kb products in 30 cycles. Reactions with iProof polymerase used a two-step protocol with acombined annealing and extension step, while the other reactions used three-step protocols with the minimum recommendedextension times. Overall reaction times include temperature ramping times.
iProof DNA polymerase amplifies long templates with high yields. Left, various fragments up to 37 kb in length were amplified from BACDNA using a combined annealing/extension step of 10 min per cycle and 30 U/ml of iProof. Right, various sequences up to 28.8 kb wereamplified directly from human genomic DNA using 30 U/ml of iProof in GC buffer with a combined annealing/extension time of 10 min per cycle.

} The events that occurred after September 11, 2001 brought the threat of bioterrorism to the forefront. One of the most important aspects of protecting against a bioterrorism attack is the ability to rapidly identify and link separate isolates of a bioterrorism agents to a common source. In the anthrax mailings of 2001, determining whether or not they were the same strain and hence from a common source was of great importance in the investigation. We used Bacillus cereus , a strain of bacteria that is closely related to Bacillus anthracis , the bacterium responsible for anthrax, as our model system. Random Amplification of Polymorphic DNA (RAPD), a type of analysis used to examine a genomic profile in organisms in which little or no genetic research has been accomplished, was used to study DNA profiles produced from different isolates of Bacillus cereus . This procedure is capable of distinguishing between Bacillus and non-Bacillus isolates and demonstrates genetic differences between strains of Bacillus cereus . Thus, this procedure can be utilized to link isolates used in bioterrorism attacks to each other and to a common source.

The genomic DNA from ten isolates of the cattle tick, Boophilus microplus collected in and around Chennai, India, was analyzed by random amplified polymorphic DNA (RAPD) using PCR. Selected five random primers were used for the study of genetic variability among different isolates of B. microplus. A high degree of genetic polymorphism with a different pattern of RAPD profiles for each tick isolate was detected with all these random primers. This variability was also confirmed by similarity coefficient values and dendrogram which were performed using mean RAPD profiles for all the primers between various isolates of ticks. The findings suggest the existence of a complex genotypic diversity of the tick B. microplus in an endemic region such as Chennai.
