Abstract To reveal the ester distribution patterns in acetylated pectins, an enzymatic fingerprinting method using a combined endo-polygalacturonase (PG) and pectin lyase (PL) treatment followed by hydrophilic interaction liquid chromatography coupled to electrospray ionization ion trap mass spectrometry with evaporative light scattering detection was developed. This method paved the way for the development of the new quantitative parameters degree of hydrolysis by PG (DHPG) and degree of hydrolysis by PL (DHPL). These parameters distinguished the methylester and acetyl group distribution patterns within different sugar beet pectins (SBPs). In the case of pectin having a degree of methylesterification (DM) of >50 and acetylation of ~20, the above approach was insufficient. Hence, a second digestion was introduced using a fungal pectin methylesterase and a PG. More than 60% of the total GalA residues present in three SBPs were recovered as monomer and oligomers after the two digestions. The first digestion of the acid extracted commercial SBP revealed the presence of small blocks of nonesterified GalA residues and segments containing large blocks of PL degradable methylesterified and /or acetylated GalA residues. Blocks of partly methylesterified, non-acetylated GalA residues were recognized after the second digestion. These results show that the acetylation pattern is non-random.

A pectin acetylesterase (BliPAE) and a pectin methylesterase (BliPME) from Bacillus licheniformis DSM13 were produced, purified and biochemically characterized. The mode of action of BliPAE and BliPME towards acetylated pectins was revealed using the newly developed enzymatic fingerprinting method. BliPAE specifically deacetylates the O-3 linked acetyl groups of nonmethylesterified galacturonic acid residues in the homogalacturan of pectin. BliPME efficiently de-methylesterifies lemon pectins (DM34-76 → DM 0) and SBPs (DM 30-73 → DM 14) in a blockwise manner. BliPME is quite tolerant towards the acetyl groups present within the SBPs. For the first time, a comprehensive experimental characterization was directed to enzymes from B. licheniformis having a PAE and a PME activity.

Foto’s digitaal aanleveren

Voor het digitaal aanleveren van foto’s dient u de af beeldingen te scannen. Scannen op 300 dpi en wegschrijven als .jpg (voorkeur) of.eps of .tiff.

- Zwart-wit foto’s bewaren als grayscale.- Kleurenfoto’s als CMYK.- Bitmap (lijntekeningen) minimaal 1200 dpi.

Het spreekt voor zich dat wij de afbeeldingen ook voor u kun nen scannen. Omslag

Als u de omslag zelf digitaal aanleverd, kunt u de in structies zoals hierboven beschreven, volgen. Vraag wel van te voren de rugdikte en lever één be-stand aan, dus achterplat, rug en voorplat com-pleet opgemaakt. Ook kunnen wij het omslag voor u verzorgen aan de hand van uw wensen. De kosten hiervan zijn variabel. +

Standard is 90 grs. matte mc paper for the interior. The reason for this is because it reads easily and is good for printing photos and illustrations. However you can choose from a variety of papers. If for in-stance you prefer a slightly tinted paper, we re-commend 90 grs. Biotop.

We print the covers on 240 grs. sulfaatkarton. You have the option of matte or glossy laminate. >>

Standaard hebben wij gekozen voor 90 grs. mat MC papier voor het binnenwerk. Dit papier is geko-zen omdat het makkelijk leest en foto’s/illustraties er goed op gedrukt kunnen worden. Maar uiter-aard kunt u kiezen uit veel soorten papier. Als u bijv. een licht getint papiersoort wilt, raden wij 90 grs. Biotop aan.

Omslag

Wij drukken de omslagen op 240 grs. Sulfaatkarton 1-zijdig gestreken. U maakt de keuze uit glanzend of mat lamineren. >>

The sizes of the cards are 10 x 15 cm and 6 x 23 cm.Usually are created to match the cover of the book, but anything is possible.

handing in the propositions

Max. two A4 papers, same design as the interior, will be reduced to 80%.Cards and theses have to be handed in at the same time as the manuscript.

Finishing

All books we produce us are paperbacks, but hard-cover is also possibility. +

Time of the production

If you design the book yourself, and deliver us the manuscript in PDF file, we can produce a good book in a relatively short time. But in order to have enough time to receive a proof and still have time for corrections, the credo is simple: the earlier you hand in your files, the more time (and ease of mind) you have to finish your book.

We need to receive your manuscript files and optional cards and theses at least three weeks before the date.

If you let us design your book, please deliver your files (and print-out) six weeks before the date. +

Benodigde tijd voor de productie

Als u de vormgeving zelf verzorgt en het manus-cript in PDF-formaat aanleverd, kunnen wij in rela-tief korte tijd een goed boek leveren. Maar om tijd genoeg te hebben voor een proef en mo gelijke cor-recties te verwerken is het credo eenvoudig: hoe eerder u inlevert, des te meer rust er is om het boek te vervaardigen.Maar in ieder geval willen we minimaal drie weken voor de inleverdatum uw zelf vervaardigde manus-cript en mogelijke uitnodigingen en stellingen ontvangen.Mocht u uw boek door ons laten opmaken, dan willen wij minimaal zes weken voor de inleverda-tum uw bestanden (met uitdraai) in ons bezit hebben. +
